To play the audio from a specific spot, hover over a line in the transcript and click the grey play button that appears to the right.

You can also 'pin' a transcript window outside of Rewind so that you can playback audio while also being able to multitask in a different app, but if you do this, the audio can only play from the beginning of the audio snippet.
